In several tissues, insulin-like growth factor I (IGF-I) is thought to act via a specific membrane receptor that interacts with the carboxyl-terminal "low affinity" binding domain of the ligand. We have identified three polypeptides with apparent Mr of 111, 116, and 130 kDa, which bind 125I-IGF-I with high affinity. These receptor polypeptides are distinct from the type I (170 kDa) and II (240 kDa) IGF-I receptor proteins previously reported. Using antisera raised to the unique carboxyl-terminal region of the IGF-I receptor, the 115-kDa (type I) and 131-kDa (type II) IGF-I receptor proteins were shown to be identical. The 115-kDa (type I) IGF-I receptor is restricted to heart and testis, and the 131-kDa (type II) IGF-I receptor is detected in heart and liver. Immunoblotting and immunofluorescence techniques reveal that both of these IGF-I receptor proteins are located on the cell surface.
